T-CellAmpTM is designed to amplify activation of human CAR-T cells, TCR-T cells, tumor infiltrating lymphocytes (TILs), and other T cell therapeutic products. T-CellAmp is added during the first 24 hours of activation. Amplification becomes apparent within 96 hours and is maintained for at least 9 days ex vivo. The effects of T-CellAmp are best seen after injection of CAR-T cells into humanized mice.

Dissolve: In 5 mL sterile PBS or basal media (no serum present). Mix vigorously to ensure complete dissolution. Store remainder solution at 4°C.
Stability: Stable for 2 weeks

This is a general protocol for using T-CellAmp. Depending on the experimental objectives, optimization may be required for maximal amplification.

Use peripheral blood monocytes (PBMCs) or T cells isolated from PBMCs.
On day 0, activate PBMCs or T cells using the activator, growth media, and expansion platform of choice. Add 4 mL of dissolved T-CellAmpTM per 100 mL growth media concurrently with the activator of choice. Scale the volume of T-CellAmp accordingly if the volume of growth media volume is less than 100 mL.
On day 1, remove growth media containing T-CellAmp and replenish with fresh growth media without T-CellAmp.
If transfection is desired, transfect cells on day 1 (after removal of T-CellAmp) or on day 2.
Follow expansion protocol of choice.
